The GeneChip® Rat ToxFX 1.0 Array consists of a single array containing 2,073 probes sets that represent 55 signatures and 22 pathways of direct toxicological interest. Analysis of the microarray data is fully automated through the ToxFX Study Builder software and ToxFX Analysis Engine, providing exhaustive documentation of the toxicological and pharmacological profile of the drug candidate or compound of interest in an easy-to-read report.
Rat ToxFX 1.0 Array
The Rat ToxFX 1.0 Array comprises a focused subset of content present on the whole genome Rat Genome 230 2.0 Array; the subset of probes represent genes associated with Drug Signatures® and pathways present in Iconix Bioscience's DrugMatrix® library.
All probe sets on the Rat ToxFX 1.0 Array are identical to their counterparts on the Rat Genome 230 2.0 Array; the focused array also includes the same hybridization, sample labeling and housekeeping controls. For a full comparison of features, please review Figure 3 and Table 1 of the data sheet.
Eleven pairs of oligonucleotide probes are used to measure the level of transcription of each gene sequence interrogated by the Rat ToxFX 1.0 Array. The oligonucleotide probes are synthesized in situ using Affymetrix' photolithograthic process, in an identical manufacturing process as the GeneChip® Rat Genome 230 2.0 Array.
ToxFX Report
The ToxFX Report is automatically generated as a PDF document and provides an in-depth summary of all aspects of the toxicology study and its results. Data are summarized at a variety of levels, from executive summary to in depth pathway analysis.
Sections of the report include:
- Executive Summary — Single-page summary highlighting key safety issues
- Study Description — Detailed record of study design
- Drug Signatures — Report on potential safety liabilities based on toxicological matches correlating with more than 50 biological endpoints (e.g., cholestasis, hepatic duct hyperplasia)
- Pathways — Detailed analysis and diagrams indicating treatment impact on 22 molecular pathways of key toxicological interest (e.g., bile acid and cholesterol metabolism or oxidative stress)
- Toxicologically Relevant Gene Groupings — A single table comprising expression data from noteworthy genes of toxicological interest (e.g., cytochrome P450)
- Methodology and Glossary — Documentation of the analytical approaches and terms required to interpret results
