Axiom Genotyping Solutionは、検証済みのSNPで構成されるAxiom Genomic Databaseからコンテンツを選択した、既製もしくはカスタムメイドのアレイプレート、必要な試薬がすべて揃った試薬キット、データ解析ツール、GeneTitan® Multi-Channel（MC） Instrument を利用して完全に自動化されたワークフローで構成されています。

Axiom Genome-Wide Arrays

ヒト疾患研究：各集団に最適化されたヒト Axiom® Genome-Wide Array Plates シリーズ。白人、アジア人、西アフリカ人集団で頻度の高いまたは低いSNPと挿入/欠損（in/del）のゲノムカバー率が最大になるように事前にデザインされています。
農業ゲノミクス：Axiom® Genome-Wide BOS1 Array Plate。　Bos taurus, Bos indicus などの肉牛および乳牛における遺伝学的、物理的SNPカバー率が最大化されています。

Axiom myDesign^TM Genotyping Arrays

完全にカスタマイズ可能な Axiom myDesign Genotyping Array Plates。 Axiom Genomic Database から選択される1,500～260万SNP、またはシーケンシングイニシアチブから取得されるお客様独自のマーカーに対応可能です。

Axiom® Genomic Database

独自に構築された生物学的よび解析的に検証済みのゲノムマーカーのデータベース、HapMap、1000 Genomes　Project、NHGRI GWAS Catalogのコンテンツを含み、Axiom myDesign プログラムのSNP選択を容易にします。

Axiom Genotyping Solution の特長

最高の遺伝的カバー率 - Axiom Genomic Database に登録されている11,000,000個の検証済みSNPによって、1000 Genomes Project、HapMap、NHGRI、などから得られる頻度の高い変異と低い変異について業界最高のカバー率を実現。各変異は、数多くの生物学的サンプルで幅広く検証されており、配列決定エラーによるものではないこと、マイナーアレルを高い信頼度で検出可能であること、Axiom アッセイで一貫して機能することが確認されています。

柔軟なフォーマット - Axiom Genomic Databaseから得られる検証済みのマーカーまたはお客様独自のターゲット配列を用いて、1,500～260万個のマーカーを搭載したアレイをデザインしていただけます。

迅速な結果 - 検証済みのマーカーおよびワークフローによって、実際のデザインとアッセイの最適化に要する時間を短縮。

高い処理能 - 自動化されたサンプル処理システムの導入と18時間以下に短縮されたハンズオン時間により、1週間あたり760サンプルを処理できます。

データの信頼度 - 事前に最適化された試薬と検証済みのワークフロー、そして経験的に実証されたSNPの組み合わせが、最高の信頼度を備えた結果を生みます。

合理化されたアッセイ

全ゲノムDNA（200ng）を増幅し、25～125塩基対(bp)の断片にランダムに断片化します。これらの断片を精製し、Axiom Genome-Wide and myDesign Array Plates にハイブリダイズさせます。ハイブリダイゼーション後、結合したターゲットを厳しい条件下で洗浄することにより、非特異的なバックグラウンドを除去し、ランダムなライゲーション反応によって生じるバックグラウンドのノイズを最小限に抑えます。各々の多型ヌクレオチドは、アレイ表面で繰り広げられるマルチカラーのライゲーション反応を通じて検出されます。ライゲーション後、アレイを染色し、GeneTitan MC Instrument 上で画像を取得します。

Axiom Genome-Wide Genotyping Assay

*試薬キットには、Beckman Biomek® FX^P Target Prep Express System でアッセイを実行するために必要となるプラスチック製の消耗品は含まれていません。必要となるBeckmanの消耗品のリストについては、Axiom 2.0 Automated Assay User Guide をご覧ください。
